Nyquist Sampling Rate calculator

This calculator helps to compute the correct sampling density (rate) for a given type of confocal laser scanning microscope and a range of imaging parameters, fulfilling the Nyquist sampling criterion.

Please choose the appropriate input values from the drop-down menus and press 'Calculate'.

The following page will return the correct zoom factor you should choose for your CLSM image acquisition. If you use this zoom factor, the CLSM software displays the correct pixel dimensions, matching the 'Sampling rate' values on the results page. The returned value for the axial sampling rate defines the Z-step interval that should be used for the Z-stack acquisition.

[1] Note the refractive indices should be matched (e.g. immersion oil lens with high RI mounting medium or water with a water corrected / dipping lens).
